AFAIK, a "Boss" is just the toughest "Named" in an instance or dungeon -- usually the furthest in and hardest to reach. There is no special (visual) indicator on the mob but they usually have the best overall loot table in the zone and are usually the objective/goal of a run.

Sample bosses:

Valdoon in the (instance) Crypt of Valdoon.

Meldrath in the (instance) Mines of Meldrath.

Varsoon in the (dungeon) Ruins of Varsoon.

Lord Ree in the (dungeon) Wailing Caves.

Not all dungeons or instances have them. I cannot think of a "Boss" for Blackburrow or Stormhold, for example.
