Can I become an Assasin if I start in Freeprort and betray to Qeynos?

I don't think so. You have to do the betrayal quest before 18, and I don't think Quynos offers the Assassin subclass. It is for that reason that you must start the betrayal quest before level 17. Once you begin the betrayal of freeport, you will be kicked out of the city, and even if you manage to sneak inside, most of the npcs will not talk to you. So if you intend to go to qeynos, you will be a ranger. Being a qeynos assassin is not possible.
